A relationship between antimicrobial peptide gene expression and capacity of a selected shrimp line to survive a Vibrio infection ArchiMer

Understanding of antimicrobial defence mechanisms of penaeid shrimp should help in the design of efficient strategies for the management and disease control in aquaculture. In this study, we have specifically analysed the expression in circulating hemocytes of antimicrobial peptides (AMPs) encoding genes, such as PEN2 and PEN3, ALF, crustin, lysozyme and a putative cysteine-rich peptide. We evidenced a relationship between the level of expression of some AMPs and the successful response of the shrimp, Litopenaeus stylirostris, to circumvent a pathogenic Vibrio penaeicida infection. A white spot disease-like syndrome in the Pacific blue shrimp (Litopenaeus stylirostris) as a form of bacterial shell disease ArchiMer

In May 1997, some white lesions evoking the white spot syndrome disease were observed in Litopenaeus stylirostris broodstock in New Caledonia. The occurrence of these lesions was neither associated with mortality, nor with histological evidence of white spot syndrome baculovirus (WSBV), The evidence suggests that these lesions result from a form of bacterial disease and are associated with an increased bacterial flora on the outer surface of the cuticle, as well as an increased incidence throughout the molt cycle. A microscopic observation of these lesions allows them to be differentiated from WSBV-associated lesions.

Acquisition of susceptibility to Vibrio penaeicida in Penaeus stylirostris postlarvae and juveniles ArchiMer

Vibriosis is a major disease problem in shrimp aquaculture, affecting all developmental stages, from larvae in hatchery tanks to juveniles and broodstock in growout ponds. However, bacterial strains responsible for vibriosis in the successive stages are usually considered to be different, and virulence specificity has been reported both at the species and at the stage levels. The New-Caledonian shrimp industry is based on the controlled reproduction of the shrimp Litopenaeus stylirostris, a species which was introduced in the 80s. The major difficulty to which the industry has been faced for 10 years is the occurrence of the "syndrome 93", which corresponds to mortality phases when the temperature falls down in April-May-June. This mortality is associated to the pathogenic bacteria Vibrio penaecida and is expressed at different levels which are variable from year ta year and from pond to pond. Arbitrarily primed PCR to type Vibrio spp. Pathogenic for shrimp ArchiMer

A molecular typing study on Vibrio strains implicated in shrimp disease outbreaks in New Caledonia and Japan was conducted by using AP-PCR (arbitrarily primed PCR). It allowed rapid identification of isolates at the genospecies level and studies of infraspecific population structures of epidemiological interest. Clusters identified within the species Vibrio penaeicida were related to their area of origin, allowing discrimination between Japanese and New Caledonian isolates, as well as between those from two different bays in New Caledonia separated by only 50 km. Shrimp aquaculture in New Caledonia is subject to seasonal mortalities during grow-out due to highly virulent Vibrio nigripulchritudo (Vn). To understand the mechanisms affecting shrimp resistance and leading to significant mortality, a shrimp ecophysiology and immunology survey was conducted on two farms, the first considered as a "control" farm (HC), the second affected by the disease (DF). Combined effect of exposure to ammonia and hypoxia on the blue shrimp Litopenaeus stylirostris survival and physiological response in relation to molt stage ArchiMer

The effect of ambient ammonia, hypoxia and combination of both on survival and the physiological and immunological response of the blue shrimp Litopenaeus stylirostris in relation to molt stage was studied. Shrimp were submitted to 44.0-71.5 mg 1(-1) total ammonia-N corresponding to 2.0 mg 1(-1) unionized ammonia NH3-N and/or to 1.5 mg O-2 1(-1) (4.3 kPa) for 24 hours. Survival was recorded and the molt stages of both dead and surviving shrimp determined. Only shrimp in intermolt and premolt stages were sampled for analysis of haemolymph. Vibrio nigripulchritudo, the etiological agent of Litopenaeus stylirostris summer syndrome, is responsible for mass mortalities of shrimp in New Caledonia. Epidemiological studies led to the suggestion that this disease is caused by an emergent group of pathogenic strains. Genomic subtractive hybridization was carried out between two isolates exhibiting low and high virulence. The influence of the soil quality of culture pond on the physiological state of Penaeus stylirostris reared in earthen shrimp pond in New Caledonia was investigated by the measurement of the osmoregulatory capacity and magnesium hemolymph concentration of samples of 20 of these shrimp captured in the pond and maintained for 24 or 48h in cages laid on the bottom. A first experiment was affected by the occurrence of a pathology caused by Vibrio penaeicida probably due to initial handling stress. The second experiment showed that the osmoregulatory capacity is depresssed as a function of the soil quality : the more it is reduced the more the depression is important.

Une substance immunostimulante administrée par voie orale est testée chez la crevette Penaeus stylirostris. Cette expérimentation est réalisée à la station de St Vincent afin de se rapprocher des conditions du Syndrôme 93. Les effets sont mesurés pendant 2 semaines sur des paramètres hématologiques (hémocytes totaux, cellules à grands granules, coloration spécifique : cellules incorporant le rouge neutre). D'autre part un stress des crevettes en cage sur un fond réduit en bassin constitue le challenge. L'administration de l'immunostimulant par voie orale augmente significativement le nombre des hémocytes totaux, ainsi que les cellules incorporant le rouge neutre. Penaeid shrimp reared in earthen ponds are exposed to sediment, which can, in some instances, induce a stress. In seawater, the osmoregulatory capacity (hyporegulation) is a useful tool to compare the physiological condition of shrimp exposed to various stressors. By keeping some shrimp in cages at different locations of a single pond heterogeneous in terms of sediment quality, it was possible, using osmotic pressure (OP), to identify some locations where the stress was maximum and some others where it was minimum. Document recommendation systems for locating relevant literature have mostly relied on methods developed a decade ago. This is largely due to the lack of a large offline gold-standard benchmark of relevant documents that cover a variety of research fields such that newly developed literature search techniques can be compared, improved and translated into practice. To overcome this bottleneck, we have established the RElevant LIterature SearcH consortium consisting of more than 1500 scientists from 84 countries, who have collectively annotated the relevance of over 180 000 PubMed-listed articles with regard to their respective seed (input) article/s. Molecular epidemiology of Vibrio nigripulchritudo, a pathogen of cultured penaeid shrimp (Litopenaeus stylirostris) in New Caledonia ArchiMer

A collection of 57 isolates of Vibrio nigripulchritudo from either diseased or healthy shrimp and from shrimp farms environment was studied in order to gain a better understanding of the epidemiology of this pathogen, notably isolated from two distinct shrimp disease complexes. Molecular typing using two different techniques, arbitrarily primed PCR (AP-PCR) and multi-locus sequence typing (MLST), studied together with experimental pathology data allowed a relevant epidemiological insight into this possibly emerging pathogen. Additionally, results obtained with the two molecular typing techniques were congruent and allowed discriminating the strains associated with the "Summer Syndrome" from strains isolated from other contexts, especially the other shrimp...
